Multixenobiotic resistance is a phenomenon in which ATP-binding cassette (ABC) family proteins transfer harmful compounds out of cells. Daphnia magna and Lumbriculus variegatus are model species in aquatic ecotoxicology, but the presence and activity of ABC proteins have not been well described in these species. The aim of this work was to study the presence, activity, and inhibition of ABC transport proteins in D. magna and L. variegatus. The presence of abcb1 and abcc transcripts in 8–9-day-old D. magna was investigated by qRT-PCR. The activity of MXR in D. magna and L. variegatus was explored by influx of the fluorescent ABC protein substrates rhodamine B and calcein-AM, with and without the model inhibitors verapamil (unspecific ABC inhibitor), reversin 205 (ABCB1 inhibitor) and MK571 (ABCC inhibitor). Juvenile D. magna possessed all examined abcb and abcc transcripts, but only reversin 205 inhibited MXR activity. The MXR activity in L. variegatus was inhibited by MK571, and to a lesser extent by verapamil, whereas reversin 205 seemed to stimulate the transport activity. Whereas calcein-AM worked better as an MXR substrate in D. magna, rhodamine B was a better substrate for L. variegatus MXR activity measurements. This is the first report on MXR activity in the order Lumbriculida, subclass Oligochaeta, and class Clitellata.

Polysyklisten aromaattisten hiilivetyjen vaikutus vesikirpun Daphnia magna ja harvasukamadon Lumbriculus variegatus monivierasaineresistenssiin
Halttunen, Teemu (2019)Polysyklisiä aromaattisia hiilivetyjä (PAH) yhdisteitä pidetään yleisesti haitallisina yhdisteinä. Joidenkin PAH-yhdisteiden on havaittu muun muassa heikentävän j eliöiden lisääntymiskykyä. PAH-yhdisteitä muodostuu ... -
Toxicity of silver nanoparticles to Lumbriculus variegatus is a function of dissolved silver and promoted by low sediment pH
Rajala, Juho; Vehniäinen, Eeva-Riikka; Väisänen, Ari; Kukkonen, Jussi (Wiley, 2018)Toxicity of silver nanoparticles (AgNPs) to benthic organisms is a major concern. The use of AgNPs in industry and consumer products leads to increasing release of AgNPs into the aquatic environment—sediments being the ... -
Toxicity of Mining-Contaminated Lake Sediments to Lumbriculus variegatus
Wallin, Jaana; Karjalainen, Juha; Väisänen, Ari; Karjalainen, Anna K. (Springer, 2021)Boreal lakes with soft water and low buffering capacity are susceptible to excess ion loading resulting from metal mining. The impact of two Finish mining sites in downstream lakes was assessed with a chronic sediment ... -
Toxicity Testing of Silver Nanoparticles in Artificial and Natural Sediments Using the Benthic Organism Lumbriculus variegatus
Rajala, Juho; Mäenpää, Kimmo; Vehniäinen, Eeva-Riikka; Väisänen, Ari; Scott-Fordsmand, Janeck James; Akkanen, Jarkko; Kukkonen, Jussi (Springer New York LLC, 2016)The increased use of silver nanoparticles (AgNP) in industrial and consumer products worldwide has resulted in their release to aquatic environments. Previous studies have mainly focused on the effects of AgNP on pelagic ... -
Behaviour of sediment-associated silver nanoparticles and their toxicity to Lumbriculus variegatus
Rajala, Juho (University of Jyväskylä, 2017)The increasing use of silver nanoparticles (AgNPs) in industry and consumer products results in the increasing environmental concentrations of AgNPs. In the aquatic environment, sediment is the major sink for the AgNPs. ...
