Fyysisen aktiivisuuden ja suorituskyvyn on useissa humaanitutkimuksissa havaittu korreloivan positiivisesti kognitioon ja aivojen terveyteen. Sama on havaittu myös hyvän ja huonon aerobisen kapasiteetin perusteella jalostetulla rottakannalla, HCR ja LCR rotilla, joita on hyödynnetty tutkittaessa perinnöllisen aerobisen kapasiteetin yhteyttä fysiologisiin muuttujiin. Aivojen dopaminergiset (DAergiset) alueet, säätelevät motorista kontrollia, oppimista ja muistia. Tämän tutkimuksen tavoitteena oli selvittää, onko perityllä aerobisella kapasiteetilla yhteyttä aivojen DAergiseen järjestelmään. Hypoteesina oli, että HCR rotat ilmentäisivät enemmän DAergisiä proteiineja verrattuna LCR rottiin jo nuorella iällä. Työssä tutkittiin dopamiinin signalointiin liittyvien proteiinien, kuten tyrosiinihydroksylaasin (TH) ja DA reseptoreiden ilmentymistä immunohistokemiallisten menetelmien avulla. ja kvantitatiivisesti analysoimalla ImageJ ja Matlab –ohjelmilla. Saatujen tulosten mukaan HCR rotilla on merkittävästi enemmän TH:n ilmentymistä striatumin alueella, vastaavasti LCR rotilla oli enemmän D1 dopamiinireseptoreita samalla alueella. Tulosten mukaan peritty synnynnäinen aerobinen kapasiteetti näyttäisi olevan yhteydessä dopamiinin tuotannon määrään striatumin alueella. Johtopäätös on, että valinta synnynnäisen aerobisen kapasiteetin perusteella luo geneettistä variaatiota myös aivojen DAergiseen systeemiin. Physical activity and fitness are shown to positively correlate with cognition and brain health in humans. Similar results have been observed in a heterogeneous rat model bred for high and low capacity running (HCR and LCR, respectively), developed to study the effects of intrinsic aerobic capacity on multiple physiological factors. Dopaminergic areas in the brain, particularly the basal ganglia, are associated with motor control, learning and memory. The Aim of this study was to investigate whether the inherited aerobic capacity is reflected in the brain dopaminergic system. Our hypothesis was that HCR rats have more dopaminergic cells or other dopaminergic proteins compared to LCR rats, already at young age. The expression of tyrosine hydroxylase (TH), dopamine receptors and other proteins involved in dopaminergic signaling, were assessed by immunohistochemical methods. Staining results were analysed by ImageJ and Matlab. The results show that HCR rats have significantly more TH expression in the striatum, whereas LCR rats have significantly more dopamine D1 receptor expression in the same area. These results indicate that together with inherited differences in intrinsic aerobic capacity, rats have inherited differences in their dopaminergic system. To conclude, the selection for intrinsic aerobic capacity seems to create genetic variation also to the brain dopaminergic system.
