dc.description.abstractPolyaromaattiset hiilivedyt (PAH) ovat ympäristössä esiintyviä orgaanisia haitta-aineita, joiden lähteitä ovat fossiilisten polttoaineiden epätäydellinen palaminen ja metsäpalot mutta myös esimerkiksi jätevedet. Reteeniä (7-isopropyyli-1-metyylifenantreeni) havaitaan erityisesti paperi- ja sellutehtaiden alapuolisten vesistöjen sedimenteissä. Tässä pro gradu -tutkielmassa tutkittiin altistuksen ajankohdan ja keston merkitystä reteenin kehitystoksisuuteen kirjolohen (Oncorhynchus mykiss) poikasissa. Kalanpoikasia altistettiin reteenille eri pituisia ajanjaksoja kahdessa eri kehitysvaiheessa ruskuaispussivaiheen aikana. Tutkittavia vasteita olivat Cyp1a-induktio, ruskuaispussitaudin oireiden ilmentyminen ja vakavuus (BSD-indeksi), poikasten pituus ja ruskuaisen pinta-ala sekä sydämen syke. Cyp1a-induktio oli selvästi koholla positiivisissa kontrolleissa, mutta merkitsevä ero havaittiin vain aikaisemmassa altistuksessa. BSD-indeksi kasvoi lähes järjestelmällisesti altistuksen keston myötä ja korreloi merkitsevästi Cyp1a-induktion kanssa. Merkitseviä eroja käsittelyjen välillä havaittiin enemmän aikaisemmassa altistuksessa. Tulosten perusteella altistuksen keston piteneminen ja aikaisempi kehitysvaihe lisäävät reteenin toksisuutta kirjolohenpoikasissa. dc.description.abstractPolycyclic aromatic hydrocarbons (PAH) are ubiquitous environmental pollutants sourcing from incomplete combustion of fossil fuels and forest fires but also e.g., industrial effluents. Retene (7-Isopropyl-1-methylphenanthrene) is found especially in the sediments downstream of paper- and pulp mills. In this master’s thesis, the effect of timing and duration of the exposure on the developmental toxicity of retene in rainbow trout (Oncorhynchus mykiss) larvae was studied. Rainbow trout larvae were exposed to retene for periods of different lengths at two time points during the yolk sac stage. The effects of the exposures were evaluated through assessing Cyp1a-induction, appearance and severity of blue sac disease (BSD) symptoms, length and yolk area of the larvae and heartbeat. Cyp1a-induction was clearly increased in the positive controls but significant difference was found solely in the early exposure. BSD-index increased almost systematically with the duration of the exposure and correlated significantly with the Cyp1a-induction. Generally, more significant differences between the treatments were found in the early exposure in comparison to the late exposure. According to the results, retene toxicity seems to increase with the length of the exposure and earlier developmental stage of the rainbow trout larvae. This is worth considering in the risk assessment of retene in the aquatic environment.en
