dc.description.abstractKirsi Ojalan väitöskirjatyön tulokset antavat lisätietoa bakuloviruksen pinnan muokkauksesta ja viruksen soveltuvuudesta geeniterapiaan. Bakulovirukset voivat tulevaisuudessa olla varteenotettava vaihtoehto kohdennettavana geeninsiirtovälineenä esimerkiksi syövän tai sydän- ja verisuonitautien hoidossa.Tutkimus osoittaa, että bakuloviruksen pinnalla voidaan tehokkaasti ilmentää vieraita molekyylejä liittämällä ne osaksi viruksen luonnollisia pintaproteiineja. Mikäli viruksen pinnalla ilmennetään solupinnan molekyyleihin sitoutuva proteiini, useimmat muokatut virukset sitoutuvat haluttuihin nisäkässolutyyppeihin. Mahdollisten geeniterapiasovelluksien kannalta onkin tärkeää, että virus pystytään sitomaan esim. syöpäsolujen pintaan ja siten siirtämään hoitogeeni haluttuun solutyyppiin.Tutkimuksessa käytetyt bakulovirukset ovat hyönteisiä infektoivia viruksia, joita on hyödynnetty jo vuosikymmenien ajan sekä luonnonmukaisessa hyönteistorjunnassa että vieraiden proteiinien tuotossa. Viime aikoina bakuloviruksia on tutkittu paljon myös erilaisissa nisäkässoluissa. Tutkimuksissa on selvinnyt, etteivät bakulovirukset lisäänny nisäkässoluissa eivätkä aiheuta ihmisille sairauksia. Ojalan tutkimuksen tarkoituksena oli kehittää uudentyyppisiä bakuloviruksia, joiden pinnalla ilmennetään erilaisia proteiineja kuten vasta-ainemolekyylejä sitovaa ZZ-proteiinia tai tiettyihin syöpämolekyyleihin sitoutuvaa vasta-ainetta. Muokattujen pintaproteiinien avulla virus pystyy tarttumaan kohdesolujen pinnalla esiintyviin vastinmolekyyleihinsä. Tutkimalla muokattuja viruksia haluttiin selvittää, voidaanko bakuloviruksia kohdentaa tietyntyyppisiin soluihin esim. syöpäsoluihin. Lisäksi tutkimuksessa testattiin, saadaanko kohdesoluissa aikaan siirrettävän geenin ilmentyminen.fi
dc.description.abstractBaculoviruses, especially Autographa calicornica multicapsid nucleo-polyhedrovirus (AcMNPV), have become a popular choice for protein expression and have been used for the production of a large variety of heterologous proteins. In addition, display of foreign peptides and proteins on the surface of baculovirus particles has raised interest as an alternative to the prokaryotic phage display system, as insect cells are able to perform complex post-translational modifications of proteins of eukaryotic origin. Moreover, the ability of baculovirus to transduce a wide range of mammalian cells has brought out the possibility to use these viruses as gene therapy vectors. The present study was aimed at the construction of efficient baculovirus display vectors that could bind to different mammalian cell types and transduce the corresponding target cells efficiently. Thus, the goal was to develop potential baculoviral tools for gene therapy applications. The foreign proteins studied were successfully incorporated on the surface of baculovirus particles and shown to be functional regarding their ligand-binding activities. The recombinant baculoviruses were further specifically bound to the membrane of the desired cells. In addition, the baculovirus expression system was applied to shed more light on the synthesis and trafficking of rubella virus (RV) envelope proteins in mammalian cells. Together, these results showed that the tropism of baculovirus could be modified, recombinant baculoviruses could be used as gene transfer vectors and expression of a marker gene in transduced mammalian target cells could be monitored. The baculovirus display and expression techniques are thus potential tools for various applications ranging from studies of protein interactions to gene therapy.en
